The Apicomplexan Parasite Toxoplasma gondii

چکیده
Toxoplasma gondii is a ubiquitous zoonotic parasite with an obligatory intracellular lifestyle. It relies on specialized set of cytoskeletal and secretory organelles for host cell invasion. When infecting its felid definitive host, T. undergoes sexual reproduction in the intestinal epithelium, producing oocysts that are excreted feces sporulate environment. In other hosts and/or tissues, multiplies by asexual reproduction. Rapidly dividing tachyzoites expand through multiple particularly nervous muscular eventually convert to slowly bradyzoites which produce tissue cysts, structures evade immune system remain infective within host. Infection normally occurs ingestion sporulated or cysts. While able infect virtually all warm-blooded animals, most infections humans asymptomatic, clinical disease occurring often immunocompromised fetuses carried seronegative mothers infected during pregnancy.

Receptor for retrograde transport in the apicomplexan parasite Toxoplasma gondii.
Toxoplasma gondii and its apicomplexan relatives (such as Plasmodium falciparum, which causes malaria) are obligate intracellular parasites that rely on sequential protein release from specialized secretory organelles for invasion and multiplication within host cells. Because of the importance of these unusual membrane trafficking pathways for drug development and comparative cell biology, char...
متن کاملModulation of the host cell proteome by the intracellular apicomplexan parasite Toxoplasma gondii.
To investigate how intracellular parasites manipulate their host cell environment at the molecular level, we undertook a quantitative proteomic study of cells following infection with the apicomplexan parasite Toxoplasma gondii. Using conventional two-dimensional electrophoresis, difference gel electrophoresis (DIGE), and mass spectrometry, we identified host proteins that were consistently mod...
متن کاملIntramembrane cleavage of microneme proteins at the surface of the apicomplexan parasite Toxoplasma gondii.
Apicomplexan parasites actively secrete proteins at their apical pole as part of the host cell invasion process. The adhesive micronemal proteins are involved in the recognition of host cell receptors. Redistribution of these receptor-ligand complexes toward the posterior pole of the parasites is powered by the actomyosin system of the parasite and is presumed to drive parasite gliding motility...
متن کاملThe Apicoplast and Endoplasmic Reticulum Cooperate in Fatty Acid Biosynthesis in the Apicomplexan Parasite Toxoplasma gondii
Background: Parasite fatty acid synthesis is an attractive drug target but complex and poorly understood. Results: We delineate the molecular activity of two pathways in Toxoplasma combining metabolomic and genetic analyses. Conclusions: The apicoplast is a significant source of fatty acids and its products are further modified in the parasite endoplasmic reticulum. Significance: We define meta...
متن کاملMultiple functionally redundant signals mediate targeting to the apicoplast in the apicomplexan parasite Toxoplasma gondii.
Most species of the protozoan phylum Apicomplexa harbor an endosymbiotic organelle--the apicoplast--acquired when an ancestral parasite engulfed a eukaryotic plastid-containing alga. Several hundred proteins are encoded in the parasite nucleus and are posttranslationally targeted to the apicoplast by a distinctive bipartite signal. The N-terminal 20 to 30 amino acids of nucleus-encoded apicopla...
